Antibody Structure Characterization
CBI has implemented a specific program for characterization of antibody structure which conforms to the following format
- Studies with the intact IgG
- Mass spectral analysis to determine molecular weight
- b. Complete amino acid analysis to determine protein content and residue equivalents per mol. This consists of 5 separate hydrolyses for quantitation of all residues, including Cys, Cystine, and Trp
- Chain separation by SDS PAGE
- a. Elution of the separated chains from the SDS PAGE gel
- Direct N-terminal sequence analysis of the separated Heavy Chain (10 cycles):
- Direct N-terminal sequence analysis of the separated Light Chain (10 cycles):
- If necessary, use of pyro-Gly aminopeptidase or Deactylase to deblock the N-terminus:
- C-terminal sequence analysis of the separated Heavy Chain
- C-terminal sequence analysis of the separated Light Chain
- Peptide mapping of the separated heavy chain.
- Peptide mapping of the separated light chain.
- Mass spec and/or automated Edman degradation primary sequence determination of individual peptide fragments from the digest of the separated heavy chain.
- Mass spec and/or automated Edman degradation primary sequence determination of individual peptide fragments from the digest of the separated light chain.
