Comparative 2D Gels

Comparative 2D Gels

Test samples are subjected to 2-dimensional (2D) electrophoresis to determine the isoelectric point of the protein and the presence of any isoforms of the protein. In the first dimension, separation is based on isolectric point in a pH gradient (generally pH 3-10), and in the second dimension, separation is based on apparent molecular weight. Proteins are visualized with Sypro Ruby Red, and stain intensities are determined by densitometry. Electrophoretograms are routinely compared using sophisticated programs which allow a statistical assessment of the "up" or "down" regulation of particular proteins visualized in the test proteome.
