Differential Scanning
Differential Scanning Calorimetry (DSC) is a method for characterizing the thermal stability of a macromolecule. The method results in determination of what is often referred to as the "melting point" of a protein or other macromolecular structure. In a DSC experiment, a solution of protein is heated (typically from 10 to 90'C) and the energy required for heating is recorded. Additional heat is needed when the protein unfolds, and this added energy appears as a peak on the thermogram. In addition to determining the temperature and molar heat of unfolding, interpretations can be made as to the mechanism of unfolding. Thus, this method of characterization is valuable in applications ranging from basic science to quality assurance.
