Protein Sequence Analysis

CBI offers two protein sequence protocols: N-terminal analysis by automated Edman degradation, and C-terminal analysis by carboxypeptidase digestion. A brief description and the prices for these services are given below:

N-terminal Analysis by automated Edman Degradation:

Starting with as little of 100 pmol of material, up to 25 residues are routinely unambiguously assigned. Quantitation of all residues is possible at the 5-10 pmol level. Many samples are provided as transblots of proteins on polyvinylidene difluoride (PVDF) membranes. With about 30 pmol of transblotted sample, as many as 15-20 residues can usually be assigned. New technologies permit direct analysis of individual protein bands excised from SDS-PAGE gels. CBI provides a transblotting and gel elution service for a an additional cost if requested. Direct sequencing of synthetic peptides still bouind to their synthesis resin is also possible.

C-terminal Analysis by Carboxypeptidase Digestion:

Starting with 1-2 nmol of protein substrate, 5-7 time point aliquots are taken, normally providing assignment of 7-10 residues. The protein sample can be provided as a solid, or in a volatile buffer such as N-morpholine acetate, pH 8.5, or 30 mM ammonium bicarbonate, pH 8.5. The concentration of the protein is critical for this technique.
