Amino Acid Analysis
Amino acid
analysis is
typically done by acid (vapor or solution phase) or base hydrolysis of protein
or peptide samples, and we also offer short time (2 hour) analyses in propionic
acid/HCl. Quantitative determination of all residues (including Trp, Pro, and
Hypro) is possible at the 800 fmol level, but the routine dynamic range is
10-100 pmoles.
In a single time point
acid hydrolysis, all residues except Cys and Trp can be quantitated. However,
to fully quantitate Ser and Thr, a second, longer time point of hydrolysis is
required.
CBI offers
reduction and alkylation (with iodoacetamide, iodoacetic acid, or 4-vinyl
pyridine) of samples for determination of Cys; free Cys can be determined
following alkylation only, and total Cys content following reduction and
alkylation. Trp is determined on yet another aliquot of sample following
hydrolysis in methanesulfonic acid. Each of these methods is considered a
separate analysis, and bears a separate cost.
