Biotechnologies has recently developed a new DNA sequencing loading buffer
(AccuTrac ®) which provides a marking tool for sample
tracking in Automated DNA sequencing applications. Specifically, use of
AccuTrac® as the sample loading buffer run on an automated DNA
sequencer produces a "mark" on the gel at the beginning of each
individual lane into which a sample was loaded. Marking the lane aids in
identification of individual lanes during the manual review of the tracking
process and ensures that the instrument accurately applies data to the
appropriate sample. AccuTrac® is detected by an automated sequencer
without interfering with the data generated by the instrument. The reagent
incorporates a fluorescent dye which is recognized by most, if not all,
currently available DNA sequencers. The formulation of
AccuTrac® is PROPRIETARY to CBI.
Below is a gel file
picture of AccuTrac ® run on a PE-Applied Biosystems 377 XL
sequencer. The DNA samples were prepared by ABI PRISM¨ BigDye®
Terminators cycle sequencing kits and post sequencing clean up was performed by
using Edge Biosystems spin columns.
The dye band at the bottom of gel enables accurate lane identification without
interfering with the data contained in the lane. Below is shown a pgem control
sequenced using AccuTrac in the loading buffer. The data shows a "GG"
base call (AccuTrac® marker) at the start of sequence data but does not
obstruct any "real data".
AccuTrac® has been
used in thousands of reactions, for both BigDye® and FS dye-terminator
reactions. It has also been used in concert with a variety of different clean
up protocols and instrumentation. Please inquire if you have any questions
regarding application of AccuTrac to your specific chemistry and
instrumentation.
